No, at least not for most car registrations.

Now, if you're new to the state, your car will need to undergo a brief vehicle identification number (VIN) inspection when you visit the county treasurer's office to get a Wyoming title or registration.

But, again, for most common vehicle registration situations, no inspection is needed; the same applies to a registration renewal.

What About a Smog Check?

Wyoming's air is fairly free of pollutants – at least compared to most states. So, the state doesn't have to force its residents to deal with car smog or emissions testing in order to comply with federal air quality standards.

That doesn't mean that emissions aren't a concern; you should do your part to reduce air pollution by performing regular maintenance on your car, limiting your trips, and buying fuel-efficient vehicles.
